Closely related to Mannophryne trinitatis according to the original publication. Barrio-Amorós, Rivas-Fuenmayor, Molina, and Kaiser, 2006, Herpetol. Rev., 37: 298-299, noted that populations from the mainland of Venezuelan formerly attributed to Mannophryne trinitatis this species were unnamed; Manzanilla, Jowers, La Marca, and García-París, 2007, Herpetol. J., 17: 32, subsequently named this species Mannophryne venezuelensis.
